Description

T-cell surface glycoprotein CD3 delta chain, also known as CD3-delta, CD3d, T3d and IMD19, is a single-pass type I transmembrane protein. CD3d contains 1 Ig-like (immunoglobulin-like) domain in the extracellular region and 1 cytoplasmic ITAM (immune tyrosine activation motif) domain. CD3d is part of the T-cell receptor (TCR)/CD3 complex and is involved in T-cell development and signal transduction. The TCR/CD3 complex of T-lymphocytes consists of either a TCR alpha/beta or TCR gamma/delta heterodimer co-expressed at the cell surface with the invariant subunits of CD3 labeled gamma, delta, epsilon, zeta, and eta. The TCR/CD3 complex plays an important role in coupling antigen recognition to several intracellular signal-transduction pathways leading to the activation of T cells. Defects in the CD3d gene cause severe combined immunodeficiency autosomal recessive T-cell-negative/B-cell-positive/NK-cell-positive (SCIDBNK)  and immunodeficiency 19 (IMD19). The genes encoding the CD3 epsilon, gamma and delta polypeptides are located in the same cluster on chromosome 11.

NCBI Gene ID: 915

Uniprot Entry: P04234

Construct Details: Full length human CD3d gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles.

Vector Type: pLTC (lentiviral expression vector containing a heterologous CMV promoter with or without a selection marker, see the vector map above)

Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)

Gene Insert Size: 516 (bp)

Primers: Gene insert coding sequence can be confirmed by following primers:

(a) forward (or 5'-end) primer: 5’-CAAATGGGCGGTAGGCGTG-3’;

(b) Reverse (or 3'-end) primer: 5’-CGGGAAGCAATAGCATGATA-3’

Note: The sequence may vary due to the naturally occurring variants or mutations, such as single nucleotide polymorphism, or the artifact during PCR amplification.  High fidelity PCR systems should always be used.
