Description

CD70, also known as CD27L (CD27 Ligand), is a member of the tumor necrosis factor superfamily (TNFSF7). CD70 is a single-pass type II transmebrane glycoprotein consisting of a short cytoplasmic domain (1-17aa), a transmembrane domain (TM: 18-38 aa) and a 155aa extracellular domain (ECD: 39-193 aa).  CD70 exists as a trimer that is found on activated T-and B-lymphocytes and mature dendritic cells. The expression of CD70 is induced by antigen receptor activation in B cells and at low levels in mouse T cells.  CD70 interacts with CD27, a member of the TNF receptor superfamily (TNFRSF7) that is expressed on natural killer (NK) cells and subsets of T and B cells. Ligation with CD27 on T cells provides co-stimulatory signals for T cell proliferation, clonal expansion and the promotion of effector T cell function. On NK cells, ligation of CD70 induces proliferation and IFN-gamma production. CD70 expression on tumor cells coupled with its highly restricted expression on normal tissues makes CD70 an attractive target for immunotherapy.

Construct Details: Full length human CD70 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles.

 

Vector Type: pLTC (lentiviral expression vector containing a heterologous CMV promoter and a marker, see the vector map above)

 

Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
