Description

CD2, also known as T-cell surface antigen T11/Leu-5 and SRBC (sheep red blood cell receptor), is a single-pass type I transmembrane protein of the immunoglobulin superfamily (IgSF). CD2 contains one Ig-like C2-type motif and one Ig-like V-type motifs in the extracellular domain (ECD). CD2 is a surface antigen of the human T-lymphocyte lineage that is expressed on all peripheral blood T cells.  CD2 is one of the earliest T-cell markers present on more than 95% of thymocytes. CD2 is also found on some natural killer (NK) cells but not on B lymphocytes. Monoclonal antibodies directed against CD2 inhibit the formation of rosettes with sheep erythrocytes, indicating that CD2 is the erythrocyte receptor or closely associated with it.  CD2 interacts with lymphocyte function-associated antigen (CD58/LFA-3) and CD48/BCM1 to mediate adhesion between T-cells and other cell types. CD2 is implicated in the triggering of T-cells and its cytoplasmic domain is implicated in the signaling function by binding to intracellular proteins. CD2 may set quantitative thresholds in T cell activation both in vitro and in vivo. The complex network of CD2-CD58 signaling also plays an important role in enhancing the adhesion of T lymphocytes to target cells, and promoting hyperplasia and activation of T lymphocytes.

Construct Details: Full length human CD2 gene (encoding UniProt #P06729) is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
