Description

CD56, also known as NCAM1 (Neural cell adhesion molecule 1) is a single pass type I transmembrane or GPI-anchored protein of the Ig superfamily (IgSF). There are 3 main forms of human CD56 that arise by alternate splicing. CD56 contains 2 fibronectin type-III motifs and 5 Ig-like C2-type motifs in the extracellular domain (ECD). CD56 may be involved in Ca++ independent cell matrix and homophilic or heterophilic cell-cell interactions.  CD56 is implicated in neural development and in neuron-neuron adhesion, neurite fasciculation, outgrowth of neurites. CD56 appears to be highly sialylated and the polysialyation reduces its adhesive property and increases its neurite outgrowth promoting features. CD56 specifically binds to heparan sulfate proteoglycans, the extracellular matrix protein agrin, and several chondroitin sulfate proteoglycans. CD56 has also been shown to be involved in the expansion of T cells, NK cells and dendritic cells which play an important role in immune surveillance.  CD56 also acts as a receptor for rabies virus.

Product Details

Gene Symbol: CD56; NCAM1; NCAM; N-CAM-1; NCAM-1; MSK39

NCBI Gene ID: 4684

Uniprot Entry: P13591

Construct Details: Full length human CD56 gene (encoding UniProt #P13591) is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter +/- a selection marker, see the vector map above)

Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)

Gene Insert Size: 2547 (bp)

Primers: Gene insert coding sequence can be confirmed by following primers:

(a) forward (or 5'-end) primer: 5’-CAAATGGGCGGTAGGCGTG-3’;

(b) reverse (or 3'-end) primer: 5’-CCAGGTTTCCGGGCCCTCAC-3’

Note: The sequence may vary due to the naturally occurring variants or mutations, such as single nucleotide polymorphism, or the artifact during PCR amplification.  High fidelity PCR systems should always be used.
