T-cell surface glycoprotein CD5, also known as Leu­1, Ly­1, and T1, is a single pass type I transmembrane protein belonging to the scavenger receptor superfamily. CD5 contains a 348 amino acid (aa) extracellular domain (ECD) with 3 scavenger receptor cysteine­rich (SRCR) domains. Crystal structure of the 3rd ECD of CD5 reveals the fold of a group B SRCR domain. The ECD of CD5 can be cleaved and circulates in the serum. T cell antigen receptor (TCR) positively regulates CD5 expression on developing thymocytes and on peripheral CD4+ T cells.  CD5 is known to inhibit TCR signaling and promotes T cell anergy and survival. CD5 signaling negatively regulates the generation of regulatory T cells (Treg) but promotes the development of Th17 cells. CD5 interacts with CD72/Lyb-2 on B cells and with PTPN6/SHP-1 for intracellular signaling. Within the B cell lineage, CD5 is expressed on B­1a cells, anergic B cells, and IL­10 producing regulatory B cells. Similar to its role on T cells, CD5 negatively regulates signaling through the B cell antigen receptor (BCR) and supports B cell survival, anergy, and tolerance. CD5 is also involved in the cellular entry of hepatitis C virus into T lymphocytes.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
