Description

LILRB1 (Leukocyte immunoglobulin like receptor B1), also known as CD85J, LIR1 (Leukocyte immunoglobulin-like receptor 1), ILT2 (Ig-like transcript 2), and MIR7 (myeloid inhibitory receptor 7), is a member of the leukocyte immunoglobulin (Ig)-like receptor (LIR) family of the Ig superfamily (IgSF).  It belongs to the subfamily B class of LIR receptors (LILRBs), which are single-pass type I trnasmembrane proteins with 2 or 4 extracellular Ig-like domains, a transmembrane domain (TM), and 2-4 cytoplasmic immunoreceptor tyrosine-based inhibitory motifs (ITIMs). LILRB1 contains 4 Ig-like C2 type domains and 4 TIIMs. The phosphorylated ITIM motif can bind the SH2 domain of several SH2-containing phosphatases to modulate cellular responses.  LILRB1 is expressed predominantly on B-cells and monocytes, and at lower levels on dendritic cells. It is also detected on a low percentage of T-cells and natural killer (NK) cells.  LILRB1 binds to MHC class I molecules on antigen-presenting cells (APCs) and transduces a negative signal that inhibits stimulation of an immune response. LILRB1 may control inflammatory responses and cytotoxicity to help focus the immune response and limit autoreactivity. LILRB1 recognizes a broad spectrum of HLA-A, HLA-B, HLA-C and HLA-G alleles. Engagement of LILRB1 present on natural killer cells or T-cells by class I MHC molecules protects the target cells from lysis. LILRB1 also binds FCER1A and FCGR1A. Interaction with HLA-B or HLA-E leads to inhibition of the signal triggered by FCER1A and inhibits serotonin release.  LILRB1 acts as a receptor for H301/UL18, a human cytomegalovirus class I MHC homolog.
