Description

LDLR (low density lipoprotein receptor) is a member of the LDLR gene family consists of cell surface proteins involved in receptor-mediated endocytosis of specific ligands. LDLR is a single-pass, type I transmembrane protein with 7 LDLR class A repeats, 3 EGF-like domains and 7 LDLR class B repeats in the extracellular region. In the cytoplasmic region, LDLR contains a NPXY motif that mediates the interaction with the clathrin adapter DAB2 and with LDLRAP1 to mediate receptor internalization. LDLR binds Low density lipoprotein (LDL) which is the major cholesterol-carrying lipoprotein of plasma.  LDL is normally bound at the cell surface to LDLR and transported into lysosomes where the protein is degraded and the cholesterol is made available for repression of microsomal enzyme 3-hydroxy-3-methylglutaryl coenzyme A (HMG CoA) reductase, the rate-limiting step in cholesterol synthesis. At the same time, a reciprocal stimulation of cholesterol ester synthesis takes place. Mutations in this gene cause the autosomal dominant disorder, familial hypercholesterolemia. LDLR also acts as a receptor for hepatitis C virus and vesicular stomatitis virus.

Construct Details: Full length human LDLR gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
