Description

PCSK9 (proprotein convertase subtilisin/kexin type 9) is a member of the subtilisin-like proprotein convertase or peptidase S8 family, which includes proteases that process protein and peptide precursors trafficking through the secretory pathway. PCSK9 undergoes autocatalytic cleavage to release the propeptide from the N-terminus and the cleavage is required for its maturation and activation. The cleaved propeptide however remains associated with the catalytic domain through non-covalent interactions, preventing potential substrates from accessing its active site. As a result, PCSK9 is secreted as an inactive protease into the extracellular matrix and trans-Golgi network. PCSK9 is expressed in liver, intestine and kidney tissues and escorts specific receptors for lysosomal degradation. PCSK9 is a crucial player in the regulation of plasma cholesterol homeostasis and fatty acid metabolism. PCSK9 binds to low-density lipid receptor family members: low density lipoprotein receptor (LDLR), very low density lipoprotein receptor (VLDLR), apolipoprotein E receptor (LRP1/APOER) and apolipoprotein receptor 2 (LRP8/APOER2), and promotes their degradation.  Mutations in the PCSK9 gene are associated with autosomal dominant familial hypercholesterolemia. PCSK9 may also regulate neuronal apoptosis and related anti-apoptotic signaling pathways

Construct Details: Full length human PCSK9 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

 

Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter +/- a selection marker, see the vector map above)

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
