Description

NTRK3 (neurotrophic receptor tyrosine kinase 3) receptor tyrosine kinase (RTK), is a single-pass type I transmembrane glycoprotein that belongs to the insulin receptor (InsR) subfamily of the tyrosine protein kinase family. NTRK3 comprises a 398-aa extracellular domain (ECD), a transmembrane region (TM), and a 386-aa intracellular kinase domain. The ECD contains 2 LRR (leucine-rich repeat) domains, 1 LRRCT and 2 Ig-like C2-type domains.  NTRK3 is involved in nervous system and probably heart development. Upon binding of its ligand NTF3/neurotrophin-3, NTRK3 autophosphorylates and activates different signaling pathways, including the phosphatidylinositol 3-kinase/AKT and the MAPK pathways, that control cell survival and differentiation. NTRK3 may play a role in the development of proprioceptive neurons that sense body position. Mutations in the NTRK3 gene have been associated with medulloblastomas, secretory breast carcinomas and other cancers.

Product Details

Gene Symbol: NTRK3; TRKC; GP145-TrkC; gp145(trkC)

NCBI Gene ID: 4916

Uniprot Entry: Q16288

Construct Details: Full length human NTRK3 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Primers: Gene insert coding sequence can be confirmed by following primers:

(a) forward (or 5'-end) primer: 5’-CAAATGGGCGGTAGGCGTG-3’;

(b) reverse (or 3'-end) primer: 5’-CCAGGTTTCCGGGCCCTCAC-3’

Note: The sequence may vary due to the naturally occurring variants or mutations, such as single nucleotide polymorphism, or the artifact during PCR amplification.  High fidelity PCR systems should always be used.
