Description

CD138, also known as SDC1 (syndecan 1), is a transmembrane (type I) heparan sulfate proteoglycan of the syndecan proteoglycan family. The syndecans mediate cell adhesion, signaling, and cytoskeletal organization and syndecan receptors are required for internalization of the HIV-1 tat protein. CD138 is a 310 amino acid (aa) single pass type I transmembrane protein with an N-terminal signal peptide, a 232-aa extracellular domain (ECD) with 6 potential glycosylation sites, a transmembrane domain (TM), and a short 35-aa cytoplasmic tail. CD138 is a cell surface proteoglycan with both heparan sulfate and chondroitin sulfate. CD138 also regulates exosome biogenesis.  CD138 shedding is enhanced by a number of factors such as heparanase, thrombin, PMA or EGF, and also by stress and wound healing. CD138 may play a role in cell proliferation, cell migration and cell-matrix interactions via its receptor for extracellular matrix proteins. Altered syndecan-1 expression has been detected in several different tumor types.

Product Details

Gene Symbol: SDC1; SDC; CD138; SYND1; syndecan

NCBI Gene ID: 6382

Uniprot Entry: P18827

Construct Details: Expressed as the combination of a heavy chain (HC) containing VH from anti-CD138 mAb and CH1-3 region of human IgG1 and a light chain (LC) encoding VL from anti-CD138 mAb and CL of human kappa light chain.  Exists as a disulfide linked dimer of the HC and LC hetero-dimer under non-reducing condition.

Source: Mammalian cells stably expressing anti-CD138 human IgG1 and growing in chemical-defined media with no animal component or antibiotics.

M.W.: Calculated molecular mass (kDa): 146 (49 kDa for a single HC, 24 kDa for a single LC); Estimated by SDS-PAGE under reducing condition: ~50 kDa (HC) + ~25 kDa (LC)

Purity: >95% judged by SDS-PAGE under reducing condition (see the gel image above, labeled as "S" and "M" for marker)

Formulation: Supplied at 0.5 mg/ml in sterile PBS pH7.4 without any carrier protein or preservative (the concentration was determined by UV absorption with an A280nm of 1.0 for 0.73 mg/ml of IgG and verified by SDS-PAGE and Coomassie blue staining).  The purified antibody was labeled with Biotin (3-5 Biotin per molecule) using the standard procedure.

Endotoxin Level: <0.1 EU per 1 μg of purified recombinant protein determined by the LAL method

Biological Activity: Binds to CD138 with high affinity (Kd < 1 nM).  Recognizes cell surface human CD138 by flow cytometry, IHC staining and fluorescent microscopy.
