RANK (receptor activator of NF­κB), also known as CD265 and ODFR, is a single-pass type I membrane glycoprotein in the tumor necrosis factor receptor superfamily (TNFRSF11A). RANK contains 4 TNFR cysteine-rich repeats in the extracellular region but lacks a cytoplasmic death domain (DD). RANK is ubiquitously expressed with high levels in skeletal muscle, thymus, liver, colon, small intestine and adrenal gland. TRANCE (TNF­related activation­induced cytokines), also known as OPGL (osteoprotegerin ligand) and ODF (osteoclast differentiation factor), is the ligand for RANK (RANKL). RANK and RANKL are important regulators of the interaction between T cells and dendritic cells for the enhancement of T cell growth and dendritic cell function. They are also essential mediators for osteoclast and lymph node development. Mutations in RANK are associated with familial expansile osteolysis (FEO), Paget disease of bone type 2 (PDB2), and osteopetrosis autosomal recessive type 7 (OPTB7). FEO is a rare autosomal dominant bone disorder characterized by focal areas of increased bone remodeling. PDB2 is a bone-remodeling disorder with clinical similarities to FEO. OPTB7 is characterized by abnormally dense bone, due to defective resorption of immature bone. Soluble RANK is able to block RANKL-induced biological activity. RANK can interact with various TRAF family proteins, through which this receptor induces the activation of NF-­κB and JNK pathways.

Construct Details: The recombinant human RANK-Fc fusion is expressed as a 411 amino acid protein consisting of Ile30 - Pro212 region of RANK (UniProt accession #Q9Y6Q6) and a C-terminal Fc from human IgG1, which exists as a dimer/tetramer/oligomer under non-reducing conditions (see the gel image inserted above).

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 3 months. The undiluted protein can be stored at 4°C for no more than 2 weeks.

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
