Description

FLT3 (fms-like tyrosine kinase 3), also known as CD135, FLK2 (fetal liver kinase 2), and STK1 (stem cell tyrosine kinase 1), is a single pass type I transmembrane protein belonging to the receptor tyrosine kinase (RTK) family and the CSF-1/PDGF receptor subfamily. FLT3 contains one immunoglobulin (Ig)-like C2-type domains in the extracellular domain (ECD) and 1 tyrosine kinase domain in the cytoplasmic region. FLT3 is detected in bone marrow, in hematopoietic stem cells, in myeloid progenitor cells and in granulocyte/macrophage progenitor cells. It is highly expressed in T-cell leukemia.  FLT3 acts as cell-surface receptor for the cytokine FLT3LG and regulates differentiation, proliferation and survival of hematopoietic progenitor cells and of dendritic cells. FLT3 is present in an inactive conformation in the absence of bound ligand and FLT3LG binding leads to homodimerization and activation by autophosphorylation. FLT3 promotes phosphorylation of SHC1 and AKT1, and activation of the downstream effectors including mTOR and RAS, leading to phosphorylation of downstream kinases, including MAPK1/ERK2 and/or MAPK3/ERK1. Mutations in FLT3 that cause constitutive kinase activity promote cell proliferation and resistance to apoptosis, resulting in acute myeloid leukemia (AML) and acute lymphoblastic leukemia (ALL).

Construct Details: Full length human FLT3 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
