Description

CD209 or CLEC4L (C-type lectin domain family 4 member L) is a single-pass type II transmembrane protein and a member of the C-type lectin superfamily.  It is also referred to as DC-SIGN (Dendritic cell-specific ICAM-3-grabbing non-integrin 1). CLEC4L is expressed on the surface of dendritic cells and macrophages. CLEC4L is involved in the innate immune system and recognizes numerous evolutionarily divergent pathogens ranging from parasites to viruses with a large impact on public health. CLEC4L is organized into three distinct domains: an N-terminal transmembrane domain, a tandem-repeat neck domain and C-type lectin carbohydrate recognition domain. The extracellular region consisting of the C-type lectin and neck domains has a dual function as a pathogen recognition receptor and a cell adhesion receptor by binding carbohydrate ligands on the surface of microbes and endogenous cells. The neck region is important for homo-oligomerization which allows the receptor to bind multivalent ligands with high avidity. Variations in the number of 23 amino acid repeats in the neck domain of CLEC4L are rare but have a significant impact on ligand binding ability. This gene is closely related in terms of both sequence and function to a neighboring gene often referred to as L-SIGN. DC-SIGN and L-SIGN differ in their ligand-binding properties and distribution

Construct Details: Full length human CD209 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
