Description

Cadherins are calcium-dependent cell adhesion proteins that preferentially interact with themselves in a homophilic manner in connecting cells.  Cadherins thus contribute to the sorting of heterogeneous cell types.  Cadherin5, CDH5 or CD144 is a 784 amino acid (aa) single pass type I transmembrane protein of the cadherin superfamily.  CDH5/CD144 contains an N-terminal signal peptide, a 552-aa extracellular domain (ECD) with 5 cadherin domains and 7 potential glycosylation sites, a transmembrane domain (TM), and a 164-aa highly conserved cytoplasmic tail.  CDH5 is found at cell-cell and cell-matrix boundaries. CDH5 can be phosphorylated on tyrosine residues by KDR/VEGFR-2 and dephosphorylated by PTPRB.  CDH5 may play an important role in endothelial junction assembly and maintenance. through control of the cohesion and organization of the intercellular junctions. CDH5 associates with alpha-catenin forming a link to the cytoskeleton. CDH5 also acts in concert with KRIT1 to establish and maintain correct endothelial cell polarity and vascular lumen. These effects are mediated by recruitment and activation of the Par polarity complex and RAP1B. CDH5 is required for activation of PRKCZ and for the localization of phosphorylated PRKCZ, PARD3, TIAM1 and RAP1B to the cell junction. The CDH5 gene is located in a gene cluster in a region of chromosome 16 involved in loss of heterozygosity events in breast and prostate cancer.

Primers: Gene insert coding sequence can be confirmed by following primers:

(a) forward (or 5'-end) primer: 5’-CAAATGGGCGGTAGGCGTG-3’;

(b) reverse (or 3'-end) primer: 5’-CCAGGTTTCCGGGCCCTCAC-3’

Note: The sequence may vary due to the naturally occurring variants or mutations, such as single nucleotide polymorphism, or the artifact during PCR amplification.  High fidelity PCR systems should always be used.
