Inactive tyrosine-protein kinase 7 or PTK7, is a single-pass type I transmembrane glycoprotein that belongs to the insulin receptor (InsR) subfamily of the tyrosine protein kinase family. PTK7 comprises a 674-aa large extracellular domain (ECD) with 7 Ig-like C2 motifs, a transmembrane region (TM), and a 345-aa intracellular kinase domain that lacks detectable catalytic kinase activity.  PTK7 is involved in the Wnt signaling pathway and plays a role in multiple cellular processes including polarity and adhesion..  MMP14 can cleave PTK7 between Pro-621 and Leu-622 generating an N-terminal soluble (70 kDa) fragment and a membrane C-terminal (50 kDa) fragment. Proteolysis by MMP14 regulates PTK7 function in non-canonical Wnt signaling pathway.  PTK7 is a component of both the non-canonical (also known as the Wnt/planar cell polarity signaling) and the canonical Wnt signaling pathway. PTK7 may function in cell adhesion, cell migration, cell polarity, proliferation, actin cytoskeleton reorganization and apoptosis. PTK7 also plays a role in embryogenesis, epithelial tissue organization and angiogenesis.

Construct Details: Full length human PTK7 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
