Description

IL10RB (interleukin-10 receptor subunit beta) is a single-pass type I transmembrane protein belonging to the type II cytokine receptor family.  IL10RB is an accessory chain essential for the active IL10 receptor complex. Coexpression of IL10RB and IL10RA proteins has been shown to be required for IL10-induced signal transduction. The IL10RB gene and three other interferon receptor genes, IFAR2, IFNAR1, and IFNGR2, form a class II cytokine receptor gene cluster located in a small region on chromosome 21. IL10RA and IL10RB mediate the immunosuppressive signal of IL10, and thus inhibits the synthesis of proinflammatory cytokines. Activation of IL10RA and IL10RB complex leads to tyrosine phosphorylation of JAK1 and TYK2 kinases. The mutations affecting the IL10RB gene are associated with inflammatory bowel disease 25 (IBD25), a chronic, relapsing inflammation of the gastrointestinal tract including Crohn disease and ulcerative colitis phenotypes.

Construct Details: Full length human IL10RB gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
