Description

CFHR1 (Complement factor H-related protein 1) is a secreted glycoprotein with 5 conserved Sushi (CCP/SCR) domains in the extracellular region. Sushi is a motif of ~60 amino acids characterized by a framework of 4 conserved half-cystine residues (1-3 and 2-4 disulfide-bonded) and several other highly conserved residues including proline, tryptophan, tyrosine/phenylalanine and cysteine. This type of structure, also designated SCR (Short Consensus Repeat), is predominantly present in proteins associated with the complement system. CFHR1 is also a member of the complement factor H (CFH) gene family, and encodes one of the 5 CFH-related (CFHR) proteins. These 5 genes are closely linked to the CFH gene on chromosome 1q31-q32. The CFHRs are secreted plasma proteins synthesized primarily by the liver and composed of highly-related SCRs. CFHR1 may be involved in complement regulation. The dimerized forms of CFHR1 have avidity for tissue-bound complement fragments and efficiently compete with the physiological complement inhibitor CFH. CFHR1 can associate with lipoproteins and may play a role in lipid metabolism.  CFHR1 binds to Pseudomonas aeruginosa elongation factor Tuf together with plasminogen, which is proteolytically activated. Tuf may act as a virulence factor by acquiring host proteins to the pathogen surface, controlling complement, and facilitating tissue invasion. Mutations in the CFHR1 gene are associated with an increased risk of atypical hemolytic-uremic syndrome.

Construct Details: Full length human CFHR1 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
