Description

BTNL2 (Butyrophilin-like protein 2) is a single pass type II transmembrane protein belonging to the BTN/MOG family of the immunoglobulin (Ig) superfamily (IgSF). Butyrophilin (BTN) is the major protein associated with fat droplets in the milk.  The BTN genes are a group of major histocompatibility complex (MHC)-associated genes that encode type I transmembrane proteins with 2 extracellular immunoglobulin (Ig) domains and an intracellular B30.2/SPRY (PRYSPRY) domain. Three subfamilies of human BTN genes are located in the MHC class I region: the single-copy BTN1A1 gene and the BTN2 (e.g., BTN2A1) and BTN3 (e.g., BNT3A1) genes, which have undergone tandem duplication, resulting in 3 copies of each.  BTNL2 contains 3 Ig-like V-type (IgV) domains in the extracelluar region (ECD).  BTNL2 is thought to be involved in immune surveillance, serving as a negative T-cell regulator by decreasing T-cell proliferation and cytokine release.  Mutations in the BTNL2 gene are associated with sarcoidosis 2 (SS2), rheumatoid arthritis (RA), ulcerative colitis (UC), inflammatory bowel disease (IBD), myositis, type 1 diabetes, systemic lupus erythematosus (SLE), acute coronary syndrome, and prostate cancer.

Construct Details: Full length human BTNL2 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter +/- an antibiotics selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
