Description

CXCR4 (C-X-C chemokine receptor type 4) is a 7-transmembrane protein of the G protein-coupled receptor 1 (GPCR1) family.  CXCR4 acts as a CXC chemokine receptor specific for stromal cell-derived factor-1 (SDF1) or CXCL12. CXCR4 transduces a signal by increasing intracellular calcium ion levels and enhancing MAPK1/MAPK3 activation. CXCR4 may also act as a receptor for extracellular ubiquitin; leading to enhanced intracellular calcium ions and reduced cellular cAMP levels. CXCR4 is involved in hematopoiesis and in cardiac ventricular septum formation. It also plays an essential role in vascularization of the gastrointestinal tract, probably by regulating vascular branching and/or remodeling processes in endothelial cells. In the central nervous system, CXCR4 may mediate hippocampal-neuron survival. CXCR4 works with the CD4 protein to support HIV entry into cells and is also highly expressed in breast cancer cells. CXCR4 promotes Env-mediated fusion of the virus. Plerixafor, an antagonist of CXCR4 activity, blocks HIV-1 entry, interaction with CXCL12 and subsequent CXCR4 signaling.  CXCR4 binds bacterial lipopolysaccharide (LPS) and mediates LPS-induced inflammatory response, including TNF secretion by monocytes.Mutations in the CXCR4 gene have been associated with WHIM (warts, hypogammaglobulinemia, infections, and myelokathexis) syndrome.

Construct Details: Full length human CXCR4 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

 

Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter +/- a selection marker, see the vector map above)

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
