Description

CD130, also known as IL6ST (interleukin 6 signal transducer), IL6RB (Interleukin-6 receptor subunit beta) and gp130, is a single pass, type I transmembrane glycoprotein of the type I cytokine receptor family, type 2 subfamily.  CD130/IL6ST consists of an extracellular region with 1 Ig (immunoglobulin)-like C2-type dpomain, 5 fibronectin type III (FN3) domains and a juxtamembrane WSXWS motif, a transmembrane domain, and a cytoplasmic tail with a box 1 motif.  The WSXWS motif is necessary for proper protein folding and efficient intracellular transport and cell-surface receptor binding, while the box 1 motif is required for JAK interaction and activation.  IL6ST/CD130 is a common signal-transducing molecule shared by many cytokines including IL6, LIF, OSM, CNTF, IL11 and CT1 which utilize IL6ST for initiating signal transmission. For example, binding of IL6 to IL6R/CD126 induces IL6ST homodimerization and formation of a high-affinity receptor complex. vIL6, a protein related to IL6 and encoded by the Kaposi sarcoma-associated herpesvirus, can bypass IL6R/CD126 and directly activate IL6ST. IL6ST mediates signals which regulate immune response, hematopoiesis, pain control and bone metabolism.  It plays a role in embryonic development and is essential for survival of motor and sensory neurons and for differentiation of astrocytes.  IL6ST is required for normal trabecular bone mass and cortical bone composition.
