Description

The human pregnancy-specific glycoproteins (PSGs) are a family of proteins that are synthesized in large amounts by placental trophoblasts and released into the maternal circulation during pregnancy.  The PSGs form a subgroup of the carcinoembryonic antigen (CEA) gene family, which belongs to the immunoglobulin superfamily (IgSF).  Members of the CEA family consist of a single N domain, with structural similarity to the Ig variable (IgV) domains, followed by a variable number of Ig constant-like 1 (IgC1) and/or 2 (IgC2) domains.  Most PSGs have an arg-gly-asp (RGD) motif, which has been shown to function as an adhesion recognition signal for several integrins, in the N-terminal domain.  PSG9 (Pregnancy-specific beta-1-glycoprotein 9) is a secreted glycoprotein of the CEA family. Human PSG9 contains 1 V-type Ig-like domain (IgV) and 3 C2-type (IgC2) domains with multiple sites for glycosylation. PSG9 is is a product of the syncytiotrophoblast.  The human placenta is a multihormonal endocrine organ that produces hormones, enzymes, and other molecules that support fetal survival and development. PSG9 is thought to inhibit platelet-fibrinogen interactions. Reduced serum concentrations of PSGs may be associated with fetal growth restrictions, while up-regulation of PSG9 has been observed in colorectal cancers.

Construct Details: Full length human PSG9 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Vector Type: pLTC or pLTC-IRES-Marker (lentiviral expression vector containing a heterologous CMV promoter, see the vector map above)

Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)

Primers: Gene insert coding sequence can be confirmed by following primers:

(a) forward (or 5'-end) primer: 5’-CAAATGGGCGGTAGGCGTG-3’;

(b) reverse (or 3'-end) primer: 5’-CCAGGTTTCCGGGCCCTCAC-3’

Note: The sequence may vary due to the naturally occurring variants or mutations, such as single nucleotide polymorphism, or the artifact during PCR amplification.  High fidelity PCR systems should always be used.
