Description

Cadherins are calcium-dependent cell adhesion proteins that preferentially interact with themselves in a homophilic manner in connecting cells.  Cadherins thus contribute to the sorting of heterogeneous cell types.  Cadherin14 or CDH15 is an 814 amino acid (aa) single pass type I transmembrane protein of the cadherin superfamily.  CDH15 contains an N-terminal signal peptide, a 546-aa extracellular domain (ECD) with 5 cadherin domains and 4 potential glycosylation sites, a transmembrane domain (TM), and a 188-aa highly conserved cytoplasmic tail. CDH15 is expressed in myoblasts and upregulated in myotubule-forming cells. CDH15 may control morphogenetic processes, specifically myogenesis, and provide a trigger for terminal muscle cell differentiation.  A chromosomal aberration involving CDH15 and KIRREL3 is found in a patient with severe mental retardation and dysmorphic facial features, termed Mental retardation, autosomal dominant 3 (MRD3).

Construct Details: Full length human CDH15 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without a selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
