CLEC4M (C-type lectin domain family 4 member C) or CD299, is a single-pass type II transmembrane protein that belongs to a the C-type lectin/C-type lectin-like domain (CTL/CTLD) superfamily. CLEC4M is often referred to as L-SIGN because of its expression in the endothelial cells of the lymph nodes and liver. CLEC4M is involved in the innate immune system and recognizes numerous evolutionarily divergent pathogens ranging from parasites to viruses. CLEC4M contains 1 tandem-repeat neck domain and 1 C-type lectin domain in the extracellular region with a dual function as a pathogen recognition receptor and a cell adhesion receptor by binding carbohydrate ligands on the surface of microbes and endogenous cells.  The neck region is important for homo-oligomerization which allows the receptor to bind multivalent ligands with high avidity.  CLEC4M is closely related in terms of both sequence and function to a neighboring gene DC-SIGN or CD209. They differ in their ligand-binding properties and distribution. CLEC4M may act as a pathogen-recognition receptor involved in peripheral immune surveillance in liver. CLEC4M may mediate the endocytosis of pathogens which are subsequently degraded in lysosomal compartments. It is also a receptor for ICAM3, probably by binding to mannose-like carbohydrates.CLEC4M acts as an attachment receptor for ebola virus, hepatitis C virus, HIV-1 ,  coronavirus 229E, cytomegalovirus/HHV-5, influenzavirus, SARS coronavirus, and west-nile virus etc.

Construct Details: Full length human CLEC4C/CD303 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter and with or without an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles. Multiple choices of a stable antibiotic selection marker are available.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
