Description

TNF (tumor necrosis factor) is the founding member of the tumor necrosis factor superfamily (TNFSF2). TNF/TNFSF2 is a single-pass type II transmebrane glycoprotein consisting of a short cytoplasmic domain (1-35 aa), a transmembrane domain (TM: 36-56 aa) and a 177 aa extracellular domain (ECD: 57-233 aa). TNF/TNFSF2 is released into the extracellular fluid, probably by cleavage from the cell surface to produce the soluble form.  TNF/TNFSF2 is mainly secreted by macrophages. TNF/TNSF2 is a multifunctional proinflammatory cytokine that binds to, and thus functions through its receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF/TNFSF2 is involved in the regulation of a wide spectrum of biological processes including cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. TNF/TNFSF2 has been implicated in a variety of diseases, including autoimmune diseases, insulin resistance, and cancer. Knockout studies in mice also suggested the neuroprotective function of this cytokine.

Primers: Gene insert coding sequence can be confirmed by following primers:

(a) forward (or 5'-end) primer: 5’-CAAATGGGCGGTAGGCGTG-3’

(b) reverse (or 3'-end) primer: 5’-CCAGGTTTCCGGGCCCTCAC-3’

Note: The sequence may vary due to the naturally occurring variants or mutations, such as single nucleotide polymorphism, or the artifact during PCR amplification.  High fidelity PCR systems should always be used.
