DR6 (death receptor 6), also known as CD358, is a single-pass, type I transmembrane glycoprotein in the TNF receptor superfamily (TNFRSF21). DR6 contains an extracellular domain (ECD) with 4 TNFR cysteine-rich repeats and a palmitylated cytoplasmic region with 1 death domain (DD). DR6 is expressed in most human tissues with the highest levels in heart, brain, placenta, pancreas, thymus, lymph node and several non-lymphoid cancer cell lines. Among hematopoietic cells, DR6 is expressed on monocytes, resting CD4+ T cells, and pro-, pre-, and naïve B cells. DR6 interacts with the adaptor protein TRADD and mediates signal transduction through its death domain. Expression of DR6 in mammalian cells induces the activation of NF-κB and JNK pathways as well as apoptosis. DR6 acts as an important regulatory molecule in T cell activation, and is involved in inflammation. DR6-defieicent mice have enhanced CD4+ T cell proliferation and are resistant to induced airway inflammation and experimental autoimmune encephalitis. DR6 is constitutively expressed on some prostate cancer cells and can be induced by TNF-α on other cytokines. DR6 is uniquely cleaved from tumor cell surface by the membrane- associated matrix metalloproteinase that is associated with tumor malignancy. DR6 is also expressed on developing neurons where it can bind APP fragment that is generated following deprivation of neurotrophic factors. APP binding to DR6 triggers DR6-mediated axonal pruning..

Construct Details: Full length human DR6 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles.

 

Vector Type: pLTC (lentiviral expression vector containing a heterologous CMV promoter, see the vector map above)

 

Formulation: Supplied with 10 μg of endotoxin-free plasmid DNA at 200 ng/μl in sterile TE buffer (pH8.0)

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
