Description

TRAIL (TNF-related apoptosis-inducing ligand) is a member of the tumor necrosis factor superfamily (TNFSF10). TRAIL/TNFSF10 is a single-pass type II transmebrane glycoprotein consisting of a 17 amino acid (aa) cytoplasmic domain, a transmembrane domain and a 243 aa extracellular domain (ECD). TRAIL/TNFSF10 may be released into the extracellular fluid, probably by cleavage form the cell surface. TRAIL/TNFSF10 is a cytokine that binds to TNFRSF10A/TRAILR1, TNFRSF10B/TRAILR2, TNFRSF10C/TRAILR3, TNFRSF10D/TRAILR4 and possibly also to TNFRSF11B/OPG. TRAIL/TNFSF10 preferentially induces apoptosis in transformed and tumor cells, but does not appear to kill normal cells although it is expressed at a significant level in most normal tissues. The activity may be modulated by binding to the decoy receptors TNFRSF10C/TRAILR3, TNFRSF10D/TRAILR4 and TNFRSF11B/OPG that cannot induce apoptosis. The binding of TRAIL/TNFSF10 to its receptors has been shown to trigger the activation of MAPK8/JNK, caspase 8, and caspase 3.

Construct Details: Full length human TRAIL/TNFSF10 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter -/+ an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months.
