Description

RANKL (receptor activator of nuclear factor kappa-B ligand) is a member of the tumor necrosis factor superfamily (TNFSF11). RANKL/TNFSF11 is a single-pass type II transmebrane glycoprotein consisting of a 47 amino acid (aa) cytoplasmic domain, a transmembrane domain and a 249 aa extracellular domain (ECD). RANKL/TNFSF11 may be released into the extracellular fluid, probably by cleavage form the cell surface. RANKL/TNFSF11 is a ligand for osteoprotegerin (OPG) and functions as a key factor for osteoclast differentiation and activation. RANKL/TNFSF11 was shown to be a dentritic cell survival factor and is involved in the regulation of T cell-dependent immune response. T cell activation was reported to induce expression of RANKL/TNFSF11 and lead to an increase of osteoclastogenesis and bone loss. RANKL/TNFSF11 may have a role in the regulation of cell apoptosis. Targeted disruption of the RANKL/TNFSF11 gene in mice led to severe osteopetrosis and a lack of osteoclasts. The deficient mice exhibited defects in early differentiation of T and B lymphocytes, and failed to form lobulo-alveolar mammary structures during pregnancy.

Construct Details: Full length human RANKL/TNFSF11 gene is subcloned into the Lentiviral expression vector pLTC with an upstream CMV promoter -/+ an antibiotic selection marker, which can be used for both transient and stable expression in mammalian cells.  It can be co-transfected with the LentiPAK DNA mix (SKU# LP-001) into HEK293 cells to produce high titer lentiviral particles.

Storage

 

The product is shipped at 4°C. Upon receipt, centrifuge the product briefly before opening the vial. It is recommended to store small aliquots at the temperature below –20°C for long-term storage and the product is stable for 6 months. 

 

 

 

 

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.
